Core Facility Resource Grant Insert Statement
The Spatial Profiling Hub Core Facility at the University of Nebraska Medical Center is a shared resource dedicated to advancing spatial multi-omics research. The Spatial Profiling Hub provides investigators with access to state-of-the-art instrumentation and expert guidance for spatially resolved molecular analysis. The facility supports multi-omic profiling of RNA and/or protein in formalin-fixed paraffin-embedded and fresh frozen tissues, enabling high-resolution spatial characterization of biological systems.
The Spatial Profiling Hub is equipped with the GeoMx® Digital Spatial Profiler, a cutting-edge platform that utilizes UV-based spatial probe collection for precise molecular profiling. This technology is well-suited for applications such as molecular subtyping of disease, biomarker discovery, cell heterogeneity analysis, and other spatially driven investigations. By offering comprehensive support from experimental design to data analysis, the Spatial Profiling Hub accelerates research in spatial transcriptomics and proteomics, facilitating novel insights into tissue architecture and disease mechanisms.

Rigor & Reproducibility Statement
The Spatial Profiling Hub ensures rigor and reproducibility by employing standardized sample preparation protocols, well-characterized antibodies/probes, and validated imaging and sequencing technologies. All immunofluorescence staining, hybridization, and washing steps followed manufacturer-recommended protocols with rigorous quality control measures, including the use of positive and negative controls. To enhance transparency, detailed metadata, including imaging conditions, reagent lot numbers, and sample processing parameters, are available upon request. All raw sample preparation protocols are documented to support reproducibility in future studies.
Data Management & Sharing Statement
Researchers are responsible for managing and safeguarding the data collected during their studies. The Spatial Profiling Hub ensures that any data generated is promptly shared with the designated researcher via their provided Box account. To enhance data security, the Spatial Profiling Hub strongly recommends that researchers create additional backups in a separate, secure location.
All data generated within the Spatial Profiling Hub will be retained for six months from the date of generation. After this period, unless otherwise requested by the researcher, the data will be permanently deleted. Researchers are encouraged to retrieve and store their data in compliance with their institution’s data retention policies.
