Molecular biology of ocular aging
Dr. Dhirendra P. Singh and colleagues study the formation of cataracts in the eye and ways to prevent oxidative damage to the lens and retina. Together with Dr. Shinohara, Dr. Singh and colleagues discovered the novel cell survival factor, lens epithelium-derived growth factor (LEDGF). LEDGF promotes the survival of many cell types, predominantly by up-regulating stress-related gene networks. Dr. Singh has also discovered that maintenance of the multifunctional protein, peroxiredoxin 6, is critical for survival of cells facing oxidative stress. This protein may be a fruitful therapeutic target for treating or postponing age-associated degenerative damage such as glaucoma, cataract, and macular degeneration. For example, a component of the spice turmeric, curcumin, prevents cataract formation by enhancing peroxiredoxin 6. Differential regulation of LEDGF and peroxiredoxin 6 may also help to explain why cells bearing identical genes respond differently to identical stresses.
